Double-stranded DMA Replication Intermediates in Human Malignant Melanoma Cells1

چکیده

DNA synthesis was investigated in human malignant mela noma cells. The DNA was extracted in a neutral nondenaturing buffer at 25°and characterized by agarose gel electrophoresis. Radioactive DNA pulse labeled for 10 to 30 min was released from the chromosomes in the form of double-stranded frag ments. The release of the fragments from the chromosomes is prevented by lysing the cells at 0°instead of at 25°suggesting that the release is enzymatic. The size of the fragments ranges between 4.6 and 5.6 x 106 daltons. After a time lag, the fragments are joined together to produce a high-molecularweight DNA.
